The most effective form of Vitamin B6

P5P
is the active form of vitamin B6. It is required for the production of neurotransmitters that control depression, pain and anxiety. Studies show that P5P has beneficial effects on depression, stress, pain, and long-term memory. It also protects nerves and reduces the symptoms of carpal tunnel syndrome, and it supports the immune system.

P5P
is the only form of vitamin B6 that can be used in the body. While some supplements use cheaper forms of vitamin B6, P5P has been shown to be the most effective for increasing plasma levels of P5P and delivering the benefits of this vitamin.

Pyridoxal 5'-phosphate (P5P) is the metabolically active coenzyme form of vitamin B6. After being consumed, B6 vitamers are metabolized into pyridoxal and then into P5P in the liver. P5P is the main circulating form and is considered the most relevant direct measure of vitamin B6 status. P5P-dependent enzymes are involved in amino acid metabolism to produce amines, many of which are important neurotransmitters and hormones, and keto-acids which are used as metabolic fuel. These enzymes are also involved in the formation of heme precursors (which are needed in blood), in phospholipid biosynthesis and in a variety of other reactions.

Vitamin B6 deficiency is associated with nervousness, depression and neuropathy. P5P is involved in the production of serotonin and GABA, neurotransmitters that control depression, pain perception and anxiety. The enzymes that produce these neurotransmitters require P5P in order to function. Therefore when P5P is deficient, the brain reduces its production of serotonin and GABA, which can lead to dysphoria as well as to neurological symptoms including tremors and seizures. P5P supplementation can increase the brain's production of both neurotransmitters.

P5P's neurological effects have many benefits. Studies in healthy older men have shown that P5P supplementation has a beneficial effect on retaining long-term memory. It has been found to help with the symptoms of premenstrual syndrome, including depression. It also works to reduce the consequences of stress. Studies have shown that P5P can prevent gastric ulcers in rats. In humans, P5P can interact with glucocorticoid receptors to down-regulate their activity, which may mitigate stress-mediated immune suppression.
P5P also has a protective effect on the nervous system. Animal studies have shown that P5P protects nerves from injury after an oxygen shortage. P5P supplementation has been shown to reduce the symptoms of carpal tunnel syndrome, in which it is normally deficient.
P5P deficiency impairs the immune system. Cellular studies have shown that P5P improves the responses of immune cells. In a human study, critically ill patients were found to have low levels of P5P despite adequate dietary intakes of vitamin B6, along with severe stress, inflammation, and clinical conditions. P5P supplementation increased the immune response of these patients.

P5P also has benefits for the cardiovascular system. Vitamin B6 is important in regulating levels of homocysteine to reduce the risk of cardiovascular disease, and two of the enzymes involved in homocysteine metabolism are P5P-dependent. Increased cardiovascular risk has been associated with low levels of plasma P5P, and its deficiency may have a role in faulty lipid metabolism. It has been found to lower blood pressure in several animal models of hypertension, and one study found that it can reduce blood pressure in humans with hypertension as well.
P5P is the only form of vitamin B that can be used in biochemical processes and has been shown to be more effective than pyridoxine hydrochloride at increasing plasma levels of P5P and delivering its benefits.

Pregnancy / Nursing: Safe at 100 mg per day

Cautions: At extremely high doses, vitamin B6 has been found to cause nerve damage. The Institute of Medicine has established a no adverse effects level (NOAEL) of 200 mg of Vitamin B6 per day, and a safe upper limit (UL) of 100 mg per day. Do not exceed these limits from all vitamin B6 supplements (pyridoxine and Pyridox-amine) combined. Discontinue vitamin B6 supplementation and consult a physician immediately if you experience neurological symptoms such as numbness, burning, pain, pricking, or tingling in your fingers or feet, or unusual clumsiness. Persons taking many drugs - including 5-fluorouracil, hydralazine, levodopa, nortriptyline, phenytoin, and tetracycline- should not take high dose vitamin B6 supplements.
